A powerful religious figure, he was responsible for spreading a deadly plague that turned an entire village into mindless automations under his control. Saddler's many underlings include village chief ], castle lord ] and super mercenary ]. He is also responsible for the creation of the several mutated creatures (such El Gigante, Del Lago and the Regenarator) that resulted from genetic experiments with the ] parasites.

Osmund Saddler is the leader of a religious cult in ] known as the "Los Iluminados" (from ], The Illuminated Ones) which seeks to overthrow the ] in order to reshape the world to their vision. He was successful in unleashing the "]" parasites which remained dormant in the Salazar family's castle by brainwashing the eighth castellan, Ramon, to do his bidding. The two contracted miners from a nearby village to excavate seal. Due to years lying dormants, the parasites have since formed into a spore-like substance, which infected the miners and later spread to the rest of the local villagers, transforming them into brain-controlled creatures known as "Ganados", or cattle(Giving some detail into the Illuminadoses's plans for them). Saddler was also capable of controlling the Ganados using a specialized sample of the Plaga vial.

In ], Saddler hired mercenary Jack Krauser to kidnap the ]'s daughter, ], as part of a plot to inject her with a Plaga egg and sent her back to the U.S. under his control under the guise of holding her for ransom.

However, Ashley's whereabouts was leaked to the U.S. by female spy ] and agent ] was sent on a rescue mission to find her. During his mission, Leon was injected with the Plaga parasite by Saddler himself while he was unconscious under captivity. This gave Saddler partial control of Leon, in addition to Ashley, as the parasites within their bodies began to harvest. He was also responsible for the death of ], a former cop posing as a hired researcher.

Leon managed to free Ashley from Saddler's clutches and the two found an Anti-Plaga Radiation Device within Saddler's island base, removing their parasites and thus, foiling Saddler's plans. Leon then fought with Saddler, after Saddler realized the Plaga had been removed from Leon, and subsequently Ashley, completely destroying his plans, and transformed into a giant four-legged creature. With Ada's help, Leon succeeded in putting an end to Saddler once and for all.
