The highway's eastern terminus was in Amarillo, Texas at an intersection with its parent, U.S. Route 66. Its western terminus was in El Paso, Texas at the U.S.-Mexico border.
The route was assigned in 1926. In 1931, US 366 was deleted in favor of an expanded U.S. Route 60 through Texas, and a rerouted U.S. Route 70 through New Mexico. The southern segment, from Alamogordo, New Mexico to El Paso, Texas would later become part of U.S. Route 54.
